Urothelial cells line the lumen bladder respond stimuli producing releasing a variety biologically active molecules. Transforming growth factor- β1 (TGF-β1), profibrotic cytokine, TGF-β receptors 1 2 are present urothelial cells. Bladder inflammation increases expression these proteins, suggesting functional role TGF-β1 both physiological pathophysiological conditions bladder. In study, we examined regulatory miR-29a on TGF-β–induced collagen primary human METHODS: Primary (HUCs) were transfected with 40 nM mimics or an inhibitor function for 24 hours. The subsequently treated (1 ng/ml) Total RNA was isolated, abundance mRNA determined real-time PCR normalized message S26, constitutively expressed ribosomal protein same sample. Collagen content cultured medium using Sircol™ assay. Data as fold change relation values (set 1) control untreated samples. RESULTS: significantly increased (5.5 ± 0.4 vs 0.1, n = 6, p <0.01). Treatment mimic reduced (0.4 0.05, <0.001 group) attenuated TGF-β1-enhanced (3.1 0.3, TGF-β1-treated group). contrast, treatment (1.8 0.2, <0.01 but did not increase TGF-β1-induced responses (6.6 0.7, 0.19 Moreover, stimulated release into culture (1.7 0.1 <0.001). abolished (p <0.01), while enhanced <0.05), effects TGF-β1. CONCLUSIONS: This is first report that regulates attenuates These results strongly suggest miR-29 may contribute maintaining regulation fibrosis disorders. Hyperglycemia- Induced NF-κB Activation Increases microRNA-146a Expression in Human Umbilical Vein Endothelial Cells

Background & objectives: Nuclear Factor kappa B (NF-κB), a master switch transcription factor, plays a critical role in the progression and development of hyperglycemia-induced microangiopathy. Hyperglycemia activates NF-κB, and subsequently increases pro-inflammatory cytokines such as TNF-α, IL-6 and IL-1β leading to development of inflammation. Some new studies have revealed the involvement o...

Skip Regulates TGF-β1-Induced Extracellular Matrix Degrading Proteases Expression in Human PC-3 Prostate Cancer Cells

Purpose. To determine whether Ski-interacting protein (SKIP) regulates TGF- β 1-stimulated expression of urokinase-type plasminogen activator (uPA), matrix metalloproteinase-9 (MMP-9), and uPA Inhibitor (PAI-1) in the androgen-independent human prostate cancer cell model. Materials and Methods. PC-3 prostate cancer cell line was used. The role of SKIP was evaluated using synthetic small interfe...

TGF-β1 regulates TGF-β1 and FGF-2 mRNA expression during fibroblast wound healing

Aims: To evaluate the expression of transforming growth factor β1 (TGF-β1) and fibroblast growth factor 2 (FGF-2) mRNA in stromal cells in response to injury in the presence of either TGF-β1 or FGF-2. It has been shown previously that heparan sulfate proteoglycans and FGF-2 are present transiently during wound repair in vivo and that an increase in TGF-β1 mRNA is detected rapidly after injury. ...

MicroRNA-124 regulates TGF-α-induced epithelial-mesenchymal transition in human prostate cancer cells.

Transforming growth factor-α (TGF-α) is upregulated in advanced stages of prostate cancer and strongly correlated with metastasis. However, the effect of TGF-α on epithelial-mesenchymal transition (EMT) in prostate cancer and the underlying mechanisms remain unclear. Recently, microRNAs have emerged as new regulators of EMT. This study found that treatment of DU145 cells with TGF-α suppressed t...

TGF-β1 Reduces miR-29a Expression to Promote Tumorigenicity and Metastasis of Cholangiocarcinoma by Targeting HDAC4

UNLABELLED Transforming growth factor β1 (TGF-β1) and miRNAs play important roles in cholangiocarcinoma progression. In this study, miR-29a level was found significantly decreased in both cholangiocarcinoma tissues and tumor cell lines. TGF-β1 reduced miR-29a expression in tumor cell lines. Furthermore, anti-miR-29a reduced the proliferation and metastasis capacity of cholangiocarcinoma cell li...
